Objetivos

The curricular unit of Separation Processes IB aims to provide unit operations of separation/purification used in biologic and related industries, providing students with the knowledge to design separation equipment.
Competences of the students:
- Identify the basic principles of the separation processes.
- Choose the most appropriate processes for a given separation.
- Design separation equipment and assess the influence of operating conditions in the separation.

Programa

1. Introduction: Stages of a separation process.
2. Movement of fluids through porous beds: Darcy's, Kozeny and Ergun Equations.
3. Fluidization: Minimum speed of fluidization.
4. Operations of Solid-Liquid Separation (suspended solids):
(i) Filtration at constant flow and constant pressure. Design filters.
(ii) Gravitational sedimentation. Sedimentation velocity; Stokes Law; Design of sedimentation tank;
(iii) Centrifuging. Stokes law applied, equivalent settling area, types of centrifuges. Equations for centrifuge’s design.
5. Solvent extraction: Extraction with organic solvents or supercritical fluids.
6. Protein Precipitation: Methods - Addition of salts, pH control in isoelectric point, addition of non-ionic polymers, addition of solvents, addition of polyelectrolyte and addition of metal ions.
7. Cell rupture: Rupture Methods - Physical, Chemical and Enzymatic Methods, and Osmotic Shock.
8. Concentration of product: Drying and freeze drying.

Métodos de ensino e atividades de aprendizagem

Theoretical material will be presented, to promote the involvement and participation of all students, developing their reasoning skills and stimulating their critical thinking. Materials about the subjects lectured will be available for consultations on Moodle. It will be expected some guided technical visits allowing for the consolidation of the themes taught. At any time students can contact the teacher via the Moodle platform.
The evaluation consists of 20% for the presentation/discussion of a scientific work and 80% for two tests or for the exam.

Fórmula de cálculo da classificação final

Evaluation in the academic year:
40% Test 1 + 40% Test 2 + 20% Work (Weight of oral presentation/discussion = 50%).
Note: there are no minimum marks. However, the weighted average of the two tests must be equal to or greater than 9.5 points in a continuous evaluation regime.

Assessment in 1st and 2nd exam season, special season exam and midterm exam:
100% Exam or 80% Exam + 20% Work (Weight of oral presentation/discussion = 50%).
Note: In exam, the minimum mark must be equal to or greater than 9.5 points.

- The Work is a seminar presented orally in a group, on a topic related to one of the unit operations taught in the curricular unit. According to the Practice-Based Research methodology, the chosen topic is directly related to a study visit to an industry or laboratory, where the group comes into contact with specific equipment(s) related to the selected unit operation. If it is not possible to carry out the study visits, the Work will be based on a scientific article.
- The student who signs the work and does not appear in the presentation/oral discussion, may be assigned a null classification in that presentation (50% weight in the final classification of the work). A different classification may be given to students from the same group based on the commitment shown, the quality of the oral presentation and the mastery of the topic.
- The Work is mandatory as a condition of continuous assessment.

Observações

In the academic year 2019/2020 the curricular unit is taught at distance.
