Objetivos

At the end of the curricular unit (UC) the student is expected to have theoretical knowledge about:

- the structure and organization of the different cellular constituents in the different types of cells;

- the function and structure of the most important biomolecules, namely, nucleic acids (RNA, DNA), carbohydrates, lipids and proteins;

- enzymatic kinetics and enzymatic assays.


They must also have theoretical knowledge and be able to apply them in a laboratory context, such as: in the preparation of typical buffer solutions for biochemical assays; in the performance of biomolecule purification techniques, namely, chromatographic and ultrafiltration techniques; determination of biomolecules (carbohydrates, proteins, nucleic acids), mastering colorimetric methods and spectrophotometric analyses; performing analytical techniques for separating proteins (acrylamide gel electrophoresis), and applying methods of analysis and determination based on in direct and indirect enzymatic assays.

Programa

Theoretical Syllabus:

1. Biodiversity of living organisms; different types of cells and their characteristics.

2. Function and structure of biomolecules: proteins, nucleic acids (DNA and RNA), carbohydrates, lipids.

3. Protein Synthesis (transcription and translation).

4. Enzyme kinetics (specific enzyme activity, Michaelis Menten kinetics, enzyme assays).





Laboratory Application Syllabus
1. Spectrophotometric methods: determination of nucleic acids or proteins by UV absorption and by quantitative colorimetric methods.

2. Preparative methods of separation of biomolecules:



  • Dialysis; centrifugation, ultracentrifugation and gradient centrifugation;

  • Filtration and ultrafiltration techniques and chromatographic methods: Hydrophobic interaction, ion exchange, affinity and molecular exclusion. Application in the isolation and purification of proteins and nucleic acids;

  • Electrophoretic methods. Separation of proteins on SDS-PAGE gel and nucleic acids on agarose gel.



3. Determination of specific enzymatic activity and kinetic parameters of Michaelis Menten. Determination of enzymatic activity and quantification methods through direct and indirect enzymatic assays.

Métodos de ensino e atividades de aprendizagem

The UC assessment is considered as a whole, integrating the theoretical and practical components. In the theoretical component, an expository-participative methodology is used, privileging the participation of students.


Theoretical-practical classes will encourage students' ability to work independently in solving exercise sheets on the themes presented in the theoretical classes. In the laboratory practical component, experimental works are developed that cover the contents of the UC and aim to provide the student with skills in the scope of "know-how to do it".


The theoretical/theoretical-practical component is evaluated through tests during the semester, and the works are evaluated through the elaboration of reports using updated and reference bibliography. The theoretical component has a weight of 50% and the practical component another 50% in the overall assessment.

Assessment in the theoretical component:

- three written tests whose average will correspond to 50% of the global assessment.

No minimum grade is imposed for each test, separately, but its weighted average must be greater than 9.5 values.


Assessment in the laboratory component, carried out through:

- Classroom context and lab notebook 10%

- Assignment 1: Individual online quizzes and discussion 20%

- Work 2: Reports of practical laboratory work 20%


The weighted average of the three classifications has a minimum score of 9.5. No minimum grade is imposed for each assessment element.

CP: 0.10 x room context +0.20 x Work 1 + 0.20 x Work 2

If the practical component is not approved, the UC cannot be approved in the current year.

Exam only for the theoretical component (TC)

100%*exam rating.

Observações

The non-validation of the laboratory component (final classification of 9.5 points) invalidates the performance of exams and approval at the UC.
