Objetivos
It is intended that students acquire skills in genetic engineering for the production of recombinant proteins in prokaryotes and eukaryotes, using several types of cloning strategies and in silico analysis. It is also intended that students develop knowledge that allow the update and progress in the area of scientific application of Molecular Biotechnology.

Programa
- Strategies for improving production of recombinant proteins. Criteria for the choice of an expression system of heterologous proteins in bacteria, and using the pET and PQE vectors. Expression systems in eukaryotes: yeast (Picchia pastoris); insect (baculovirus); mammals; viral vectors for expression in mammalian cells; promoters used in the construction of vectors for mammalian cells; for plants with Agrobacterium.
- Methodologies to promote genetic fusion and genetic interruption: "one step disruption method" and "PCR-generated short and long flanking homology". Primer design for fusion, disruption, directed mutagenesis and gap repair. Cloning strategies. Gene regulation by RNA Interference.
- In silico analysis of sequences. Nucleotidic sequence searches. Alignment of DNA or protein sequences. In silico analysis of the topology of a protein. Phylogenetic analysis of proteins.
- Fundamentals of microbial evolution.

This curricular unit comprises a theoretical/practical component. The theoretical contents are exposed via a computer readable presentations of PowerPoint and are accompanied by exercises of practical application that includes classes for the analysis of case studies and mini-projects that can be partially tested in the laboratory, in the Practical Laboratorial component of the CU.
