Objetivos

At the end of this course it is intended that the student:
- Understand the role of genes as the molecular basis of the mechanisms of genetic alteration and its fundamental importance for the establishment of recombinant DNA technology;
- master Molecular Biology and Genetic Engineering methodologies that are the basis for genetic analysis and genetic research at the molecular level, namely identification of serotypes, genotypes, etc.;
- to apply recombinant DNA techniques for cloning in microrganisms.

Theoretical component:
1. Introduction to Genetic Engineering.
2. Recombinant DNA technology, role of restriction enzymes and vectors.
3. Nucleic acid visualization and quantification methods: agarose gel electrophoresis, Southern and Northern blot.
4. Cloning of genes.
5. Expression systems.
6. DNA libraries.
7. Amplification of nucleic acids by PCR (qualitative and quantitative techniques: qPCR, RT-PCR, Real Time-PCR).
8. Sequencing Methods
9. gene therapy

Laboratory Application Component:
1. Nucleic acid extraction (genomic DNA, plasmid DNA and RNA)
2. Analytical and quantitative agarose gel electrophoresis;
3. Digestion of DNA with specific enzymes – restriction pattern;
4. PCR amplification of DNA samples;
5. Cloning of genes in expression vectors;
6. Recovery of DNA samples from agarose gel.

Métodos de ensino e atividades de aprendizagem

The contents of the UC are presented in lectures, in expository mode interspersed with examples, using the support of powerpoint slides and in the current academic year, in online mode through the Zoom platform.

The laboratory component allows the practical illustration of theoretical concepts. In laboratory classes, 3 large works are performed, covering more than 1 class/week, favoring the strategy of combining the illustration and execution of the various techniques described in the syllabus, for a specific objective.
Students are integrated into an interdisciplinary research project that involves other Graduate courses. The products (purified genetic material, and/or prepared with modifications) obtained in their work are used as raw material by the other students, who produce other recombinant genetic material which in turn serves as a sample to be analyzed in one of the last laboratory works. Works are developed that illustrate the most relevant laboratory techniques and the interpretation of results and problem solving. In the current academic year, laboratory classes may be concentrated in modules of 4 to 6 hours.

Fórmula de cálculo da classificação final

Continuous Assessment:

FG = 50 % (theoretical component: 2 tests) + 50% (Laboratorial component)

Assessment by Exam*:

FG = 50% Exam + 50% (Laboratorial component)

CU with SPECIAL REGIME (100% eval. by exam not possible) – ONLY THE THEORETICAL COMPONENT CAN BE EVALUATED BY FINAL EXAM.

Provas e trabalhos especiais

The laboratory component is evaluated by 3 parameters:


  1. Laboratory work reports: 40%

  2. Preparation, behavior and participation in laboratory execution: 50%

  3. Preparation and maintenance of laboratory notebook: 10%.



An oral evaluation / discussion of the reports will be carried out. Members of the same group may obtain different grades.

Avaliação especial (TE, DA, ...)

There is no provision for any special assessment regime, not included in those already provided for in the Regulation.
