Objetivos

This unit present a strong practical-laboratory component, promoting students to achieve skills for laboratory work autonomy, as well as to plan the experimental work according to the theoretic basis of the studies. The collaboration with the Faculty ofMedical Sciences of the New University of Lisbon, in particular with the CEDOC laboratories, is an added value to this unit.

This unit has as its primary goals:1. To develop an integrated perspective of the techniques used in molecular genetics for diagnostics or research, bas in four processes: amplification, separation, detection, and expression; 2. To understand the theoretical basis of the techniques of molecular genetics;3. To learn common techniques of molecular genetics.

Theoretical:1. Organizational structure of a genome2. Genome sequencing techniques3. Genetic expression analysis at the genome level: transcriptomics and proteomics. Experimental methods used to study the regulation of genetic expression4. The analysis of genetic expression at the gene level: genetics screenings5. Techniques of extraction of nucleic acids6. Techniques of amplification of nucleic acids (PCR, Real time, etc.)7. Techniques of sequencing and analysis of nucleic acids8. Mapping, cloning and genetic expression9. Molecular genetics techniques using bacteria and phages
Laboratory:1. Rules for laboratory safety;2. Preparation of solutions and pipetting;3. Basic techniques of manipulation of nuclei acids: DNA extraction, gel electrophoresis4. DNA amplification: PCR and Real time PCR5. Analysis of DNA sequences6. Bacterial cloning: construction of recombinant molecules.

Métodos de ensino e atividades de aprendizagem

The teaching methodologies are based on expository and participatory methods using virtual collaborative platforms, to promote discussion and analysis of procedures and results. In the theoretical component, contents will be presented using digitalplatforms and assuming slides projection, films and animations visualizations. The practical component ensure laboratory classes with students following protocols for different works. The e- distance teaching, includes synchronous and asynchronousactivities.

Assessment methodology: continuous, consisting of report (60%) about laboratory works, performed as group work (3-4 students) and an individual written test (40%).The student will be able to do final assessment, by an exam consisting of a written test with contents of the theoretical and laboratory components, accounting for100% of the UC evaluation.
