Objetivos

At the end of the curricular unit (CU), the student is expected to have theoretical knowledge about: - the structure and organization of different cell constituents in different types of cells; - the function and structure of the most important biomolecules, namely nucleic acids (RNA, DNA), carbohydrates, lipids and proteins; - have notions of enzymatic kinetics and enzymatic assays.


More than acquired the theoretical knowledge students should know how to apply it in a laboratory context: - preparation of buffer solutions typical of biochemical assays, - biomolecule purification techniques, namely, chromatographic and ultrafiltration techniques, - biomolecules assay techniques (glycosides, proteins, nucleic acids ), mastering colorimetric methods and spectrophotometric analysis, - analytical techniques for protein separation (acrylamide gel electrophoresis), - analysis and assay methods based on direct and indirect enzymatic assays.

Theoretical Contents:



  1. Biodiversity of living organisms; different types of cells and their characteristics

  2. Function and structure of biomolecules: proteins, nucleic acids (DNA and RNA), carbohydrates, lipids.

  3. Protein Synthesis (transcription and translation)

  4. Enzyme kinetics (specific enzymatic activity, Michaelis Menten kinetics, enzymatic assays)


Laboratory Application Contents


  1. Spectrophotometric methods: determination of nucleic acids or proteins by UV absorption and by quantitative colorimetric methods.

  2. Preparative methods of separating biomolecules:



  • Dialysis; centrifugation, ultracentrifugation and gradient centrifugation.

  • Filtration and ultrafiltration techniques and chromatographic methods: Hydrophobic, ion exchange, affinity and molecular exclusion interactions. Application in the isolation and purification of proteins and nucleic acids.

  • Electrophoretic methods. Separation of proteins on an SDS-PAGE gel and nucleic acids on an agarose gel.


3. Determination of specific enzymatic activity and kinetic parameters of Michaelis Menten. Determination of enzymatic activity and quantification methods through direct and indirect enzymatic assays

The CU assessment is considered as a whole, integrating the theoretical and practical component. In the theoretical component, an expository-participatory methodology is used, prioritizing student participation. Theoretical-practical classes will allow to foster the students' ability to work independently in the resolution of exercise sheets on the themes presented in the theoretical classes. In the laboratory practical component, experimental works are developed that cover the contents of the UC and aim to provide the student with skills in the scope of “know-how”. The theoretical/theoretical-practical component is evaluated through tests during the semester, and the works are evaluated through the elaboration of reports using updated bibliography and reference. The theoretical component has a weight of 50% and the practical component another 50% in the global assessment.

Assessment in the theoretical component:


Average of the classification obtained in the three written tests carried out throughout the semester


No minimum score is imposed for each test separately, however its weighted average must be equal or greater than 9.5.


Evaluation in the laboratory component:,

Carried out through:


- Classroom context and laboratory notebook 10%


- Assignment 1: verage of the classification obtained in the complete reports of practical laboratory work 25%


- Assignment 2: Discussion of the reports 15%


The weighted average of the three classifications has a minimum score of 9.5. No minimum grade is imposed for each assessment element

Practical Component (PC)

PC: 0.10 x classroom context +0.25 x Assignment 1 + 0.15 x Assignment 2

The non-approval of the practical component makes it impossible to approve the CU in the current year

Exam only for the theoretical component (TC)

Observações

The tests performed in the theoretical-practical component do not have a minimum grade, but their average must be equal to or greater than 9.5.


The non-validation of the laboratory component (final classification of 9.5) invalidates the performance of exams and approval in the CU.
